Rutgers researchers may have stopped HIV

Piscataway, N.J. — Researchers at Rutgers University have developed a trio of drugs they believe can destroy HIV, the virus that causes AIDS, according to a published report.

The drugs, called DAPYs, mimic the virus by changing shape, which enables them to interfere with the way HIV attacks the immune system.

Tests conducted in conjunction with Johnson and Johnson have shown the drug to be easily absorbed with minimal side effects. It also can be taken in one pill, in contrast to the drug cocktails currently taken by many AIDS patients.

“This could be it,� Stephen Smith, the head of the department of infectious diseases at Saint Michael's Medical Center in Newark, said. “We're all looking for the next class of drugs.�

A research team led by Rutgers chemist Eddy Arnold pre-published details of the most promising of the three drugs, known as R278474, last month in the electronic edition of the Journal of Medicinal Chemistry. Full details will be published in the journal in early 2005.

Dr. Arnold, 47, has worked at dismantling the AIDS virus over the last 20 years. He uses X-ray crystallography, a technique to determine the structure of molecules, the smallest particles that can retain all the characteristics of an element or compound.

The research has targeted reverse transcriptase, a submiscroscopic protein composed of two coiled chains of amino acids. It is considered HIV's key protein.

“Reverse transcriptase is very important in the biology of AIDS,� Dr. Smith said. “If you can really inhibit reverse transcriptase, you can stop AIDS.�

The optimism about R278474 stems from its potential to interfere with an enzyme that the virus needs to copy and insert itself into a human cell.

“We're onto something very, very special,� Dr. Arnold said.

Dr. Arnold established his lab at Rutgers' Center for Advanced Biotechnology and Medicine in 1987. His current 30-member research team is partnered with Johnson and Johnson subsidiaries Janssen Pharmaceutica and Tibotec-Virco NV.

An important advancement in Dr. Arnold's research came in 1990 when Belgian scientist Paul Janssen was added to the collaboration. Dr. Janssen, considered a drug pioneer, published a paper that year that described a new drug that blocked reverse transcriptase but caused resistant strains of the virus to pop up too quickly.

Dr. Janssen sought out Dr. Arnold, who used crystallography to detail the structure of RT. Their work ultimately led to the RT inhibitors.

“We may eventually win the war against HIV/AIDS. That would be an extremely rewarding and satisfying outcome,� Dr. Arnold said.

jaross, first off, do you have any idea how much it costs to make any new drug, let alone one like this? theres a reason that drugs are pricey. on average it costs $800,000,000 to bring any new drug on the market. some are more and some are less. im guessing this AIDs drug was probably more. if you want to have this country adopt a more canadian like perscription drug policy, then you can kiss drugs like this goodbye. because you know what? pharma companies have to turn a profit if theyre going to keep putting out new products. so can it.

reverse transcriptase inhibitors have been around for a long time. This is how AIDS works...the virus gets into your cells and then inserts its genome into YOUR DNA. Once its in, there is absolutly no way to get it out, besides killing the infected cell. Once inserted into your DNA it can just chill out for a while and do nothing, or maybe after a while decide to use YOUR DNA REPLICATION MECHANISMS to produce HIV virus protenis that eventually assemble into complete virus particles, which are released from that cell and go on to infect others. Reverse transcriptase inhibitors work because they prevent the HIV RNA from being converted to DNA (only when its converted to DNA can it insert into your DNA). SO really...it only prevents HIV spread, and does nothing to get the HIV DNA out of your cell that is allready there. ANY CELL THATS INFECTED WITH HIV HAS IT FOREVER...when that cell divides, it it will also has the HIV gene and can produce new HIV viruses. RT inhibitors may be able to keep the virus under contoll, but cant get rid of it. you have to kill infected cells to get rid of it. If infected cells include the vast majority of your body...then your in trouble. Sounds like this new drug can stop HIV integreation into uninfected cells, but a cell that is already infected it will continue to produce new virus that will have to be dealt with by the immune system, and depending on the levels of infection, RT may or may not help you get better. They do work though, and increase lifespan dramatically...just wont completly cure you.

oh ya...and herpes is troublesome because it hides out in neurons in the brain, where the immune system cells never get to. I guess you could make a drug to kill herpes infected neurons...but depending on how much you use those nerurons in the first place...you may not want to do that.

i dont see how the ideas are 'dumbass' for two reasons. fist, they arent my ideas per se. i was talking about an article in newsweek that i read about it, so take up your differences with howard fineman, not me.

second, yes, i know that one of the largest pharma manufacturers, merck has plants in and out of the US. so what? lots of US companies have plants outside the country. their earnings still go into the US GNP.

the reason that you can get american drugs cheaper in countries like Canada, and i am assuming Slovakia, is because there are government controlls on how high the price can be. this is how things are in a socialized nation. youre paying the extra in your sales tax.

in america, there is now price ceiling on products generally, and this is certainly the case for the drug companies. they sell not only to american consumers at market prices, but they can also sell to the national governments of other countries, at whatever price is agreed on. the govenments of those countries then affix whatever price they think is beneficial for their country, and make them available for that price.

so, the problem here is that if american comsumers run across the boarder to buy drugs at socialist prices, the market in the US will fall flat (this is hypothetical) as i mentioned before, if no one is going to buy the drugs at the US market price, then the price has to come down right? right. so what happens then, is that Merck and pfizer, and other manufacturers bring in less profit. as the newsweek article mentioned, it costs roughly $800,000,000 in r&d to bring a new drug on the market, due to FDA standards, and other mandated tests. that is how we keep debachles like the one that merck just went through to to a minimum.

so, if the drug companies arent turning as large of a profit anymore, due to governmental price ceilings, where are they going to come up with the 800 million dollars to bring out new products like the one mentioned in this thread? well, the answer is, either the government, or nowhere. clearly the private sector is going to produce greater results than another government department with all kinds of buerocracies to entangle and slow things down.

really the proof is in the results. you can look at which nations are the most productive, and which have contributed more to the world than others. socialism is a recipit for mediocrity. this is just another example of that.

roadgapwhatever ure name is, you basically just described a virus. HIV is special though, it goes right after your immune system, destroying your ability to make white blood cells and platelets. Once this has happenned, a common cold can kill you, or even cutting yourself shaving. Because you have no platelets, you cant heal properly and that allows even more pathogens to get into your body.

However, the cure they are proposing, unless i am incorrect, stops HIV from causing AIDS. You can still be a carrier of HIV, and if HIV has allready mutated into AIDS, that damage is irreversible.

So what we have here, is a genetically modified protein, which acts just like a virus to stop a virus from reproducing. Essentially, we have created a true 'anti-virus', there is no way to be able to predict exactly what happens in the long run, it could mutate itself, and pose an even bigger problem(hell, we could even create an airborn HIV strand, say goodbye to most of humanity or even worse, turns people into zombies). We have no idea what we are dealing with, and without at least another 10 years of research, and then 10 years before it gets onto the market. In 20 years, a substantial part of africa, asia and parts of europe will have a considerably tough time fighting aids. I know it sounds pessimistic and very far fetched, but i really dont see a real cure on the horizon.

anewmorning, you have said some of the most ridicilous things. Drug companys put next to no money into solving things like HIV and cancer. almost all of their research goes into cold medication. Second, canadians pay the same for their drugs. I dont get free precription drugs when im sick, i have to pay for everything. Just my taxes go and help pay for it, so i dont pay as much. I dont know alot about the economics of it, but i do know that drug companies spend most of their money on medications which make us feel comfortable and have nothing to do with saving lives.
