GOD that's a tough question. I mean just out of LOTR it's almost impossible. Higgins from Pygmalion, Henry Wotton from the Picture of Dorian Grey, Hamlet, Iago, Don Quixote, Heathcliffe from Wuthering Heights, Captain Ahab... There are just way too many characters out there to pick one and say he or she is the 'best'.

But I will anyways. Samwise Gamgee. Aragorn was the true protagonist of LOTR, but Sam was the hero. Like several other characters, the movies really didn't do him justice.

I am saddened by the number of people here who don't read books. Sorry, but the greatest fictional character of all time will not be found in a movie (not even an adaptation; they always fail to get the whole character across). You simply don't have time in 2 hours to bring across a complete character, much less a great one, in comparison to a book or a play. It's amazing Shakespeare manages to do it with plays, actually, but hey, that's why he's the Bard. Everyone who said Caufield has a point, though, he's a decent pick.
